Career / Scientific Context / Missions

After a PhD in Immunology, I was recruited in 2002 to lead projects combining cell biology, molecular biology and biochemistry. In 2011, I joined Laurent Groc’s team during IINS Institute creation.

The major interest of our team is to understand the excitatory synapse physiology, fundamental for learning and memory processes, and the pathological dysfunction in psychiatric disorders. In particular, we are studying a central receptor of the excitatory synaptic transmission, the NMDA-type glutamate receptor (RNMDA), its cell trafficking using cell biology and high-resolution microscopy techniques, and its dysfunction induced by immune mechanisms in psychotic disorders.

My functions:

1/ Technical coordination of the research team and supervision of a technical team producing cellular and molecular biology tools (primary cultures, organotypic slices ; Plasmids and recombinant viruses constructs); Transversal activities for the Institute : Health and Safety preventing officer, Founding member of the cell culture Facility.

2/ Conducting a translational research project on the identification of autoantibody targets in psychotic patients with biochemical approaches: analysis of protein-protein interactions with co-immunoprecipitation, PLA approaches (TurboID); Developing and technically leading a project of multi-approach diagnosis platform between our team and Bordeaux University Hospital. Our aim is to create a multi-step diagnosis, to detect autoimmunity in psychotic disorders, adapted to clinical practice and define the good process to detect the molecular pathogenicity of patients autoantibodies given that the single detection of antibodies is not a marker of pathology. As technical manager, I coordinate and structure the design phase and the implementation of the differents experiments by supervising the dedicated staff. This project is supported by SPARK fundings for 2 years and others grants (Fondation FondaMental, CNRS, UB).

- Cell biology: Hippocampal and cortical primary cultures, organotypic rodents Cultures, cell lines, Exogen DNA expression after transfection, electroporation, viral transduction, immunolabelling.

- Biochemical approaches: western-blot, surface biotynilation, co-immunoprecipitation, synaptic purification after subcellular fractionation, BioID PLA assay

- Molecular biology: DNA constructs strategies with ligation, restriction, PCR, Directed mutagenesis, in Fusion
